Lymphoma and hemangiosarcoma are both aggressive cancers with poor prognosis. Dogs also develop lymphoma and hemangiosarcoma and make an excellent model system for both diseases. Research shows that dogs and humans have similar genetic predisposition for many of the diseases where genetic background and environment play a role, such as several forms of cancer. The dog population structure, with distinct breeds derived from small founder populations, together with pedigrees, makes trait mapping easier than in humans. The purpose of this study is to identify inherited risk factors, as well as somatic mutations commonly seen in lymphomas and hemangiosarcomas, two cancer types that have a high incidence in e.g. golden retrievers. Especially lymphomas are among the most clinically and histopathologically diverse of all human cancers, making diagnosis and clinical management difficult. We aim to identify and characterise germ-line and somatic mutations that impact the development and progression of lymphoma and hemangiosarcoma in high-risk breeds, and use this to find relevant mutations and chromosomal changes in humans. We have promising preliminary findings with high correspondence with human data. Finding molecular markers will allow a comprehensive subclassification of distinct tumour subtypes, enabling the development of therapies targeted to specific mechanistic pathways. We also aim at developing a personalised vaccine against lymphoma.
