Recent work shows that growth factor receptor signalling continues in endosomes, turning the endosomal network into a signalling platform. Different growth factors modulate the endosomal network and features like endosome number and size can affect cell fate decisions. However, it is unknown whether this mechanism has relevance at the organ level in vivo. The proposed project aims to clarify the role of the endosomal system in the regulation of signalling in the adult and regenerating mouse liver, using a combination of high-resolution microscopy, quantitative image analysis and state-of-the-art RNA interference technology. First, I will analyze the effects of HGF and TGF beta, two growth factors leading to distinct cell fate decisions, on the organization of the endosomal network in hepatocytes in vitro. Second, I will evaluate how endosome distribution changes in response to signalling during liver regeneration after surgical removal of a part of the liver.
