Beskrivning
Forskningsgruppen Farmakologi studerar transportörer från Solute Carrier (SLC) familjen och deras roll i cellulär metabolism, i upptag av läkemedel och gifter och deras potential som framtida läkemedelsmål. Det finns över 400 SLCer hos människa, med uppgift att transportera näringsämnen, socker, aminosyror, läkemedel och andra substrat över plasmamembranet och även över membranet på de flesta av cellens organeller. Omkring 30% av alla SLCer har inga kända substrat som de kan transportera. Ett av våra huvudfocus är att identifiera molekyler som dessa kan transportera. Vi gör detta med olika metoder, bland annat Thermal Shift, upptagsförsök samt elektrofysiologiska mätningar i Xenopus laevis oocyter och i eukrayota celler. Ett annat huvudfocus för vår forskning är att identifiera funktionen för de olika SLCerna i cellens och hela organismens fysiologi. För detta använder vi genutslagning i möss och bananfluga Drosophila melanogaster, samt CRISPR/CAS editering av gener i cell-linjer, primära celler från mus och stamceller. Vi använder också storskaliga metoder så som RNA sekvensering, microarrays, proteomik, high content imaging och metabolomik. Vårt slutgiltiga mål är att identifiera vilka kombinationer av transportörer som är uttryckta på olika celler och vilka SLCer som är ansvariga för upptaget av ett givet substrat på respektive celltyp. Med den kunskapen kan vi identifiera läkemedelsmåltavlor för att aktivera eller stänga av olika system i cellerna samt att styra upptaget av läkemedel in i vissa specifika celltyper.
