Astrocyters roll för Alzheimers och Parkinsons sjukdom
Målet med vår forskning är att förstå hur hjärnans astrocyter bidrar till sjukdomsutveckling och spridning av patologin vid Alzheimers och Parkinsons sjukdom. Genom denna nya infallsvinkel hoppas vi kunna identifiera framtida behandlingsstrategier. Forskningen på området har under många år varit inriktad på förändringar som sker i nervcellerna, men alltmer uppmärksamhet riktas nu mot hjärnans olika stödjeceller.

Förändringar i den sjuka hjärnan
Både vid Alzheimers och Parkinsons sjukdom sker en ansamling av skadliga proteiner. Amyloid-beta och tau bildar stora klumpar i Alzheimer-hjärnan, medan aggregat av alfa-synuklein ansamlas i Parkinson-hjärnan. Dessutom pågår en omfattande inflammation, som ökar under sjukdomsförloppet. Trots att man länge vetat om dessa förändringar, återstår många frågetecken kring mekanismerna som gör att sjukdomen förvärras och sprids till nya regioner av hjärnan.
Astrocyter sprider sjukliga proteinaggregat
Astrocyterna är den absolut vanligaste typen av stödjecell och har mycket stor inverkan på hjärnans hälsa. Vår forskning visar att astrocyter tar upp stora mängder aggregerat amyloid-beta, tau och alfa-synuklein, men att de är mycket dåliga på att bryta ner det intagna materialet, vilket i stället ackumuleras inuti cellerna. Detta orsakar allvarlig cellulär stress och leder till att det intagna proteinet sprids till närliggande celler med olika mekanismer. Till exempel kontaktar astrocyterna sina grannar med tunna trådlika utskott (nanorör), samt släpper ifrån sig vesiklar med delvis nedbrutet material, som skadar omgivande nervceller. Våra resultat indikerar att astrocyter kan vara en måltavla för att bromsa sjukdomsprocessen.
Metoder
För att undersöka hur astrocyter påverkar olika patologiska processer har vi utvecklat användbara sjukdomsmodeller, baserade på humana iPSC-deriverade astrocyter, nervceller och mikroglia. Med hjälp av ett batteri av olika tekniker, såsom live cell imaging, flödescytometri och immunofärgningar, samt protein och lipidanalyser kartlägger vi astrocytmedierade sjukdomsmekanismer. Vi använder även 3D organoidkulturer och humanvävnad i flera av våra projekt.
