Långtidsminnets mekanismer, funktioner hos genfamiljer, och egenskaperna hos G-proteinkopplade receptorer.
Min grupp har tre huvudsakliga forskningsprojekt:
1) Att reda ut evolutionen för långtidsminnes mekanismer. Det molekylära maskineriet för att skapa minnen utgör en av de mest utmanande frågorna för neurobiologer att klarlägga. Dessa komplexa mekanismer kan förstås endast genom att studera deras komponenter. Vår strategi är att undersöka hur och när generna och deras proteiner uppstod som är inblandade i dessa processer. Vi vet redan att de flesta generna uppstod i samband med ryggradsdjurens uppkomst för mer än 500 miljoner år sedan. Vi använder zebrafisk som modelldjur för att reda ut minnesproteinernas utbredning och funktioner.
2) Att reda ut evolutionen för viktiga genfamiljer hos ryggradsdjur, särskilt genfamiljer som är uttryckta i nervsystemet och i det endokrina systemet. Syftet är att klarlägga vid vilka tidpunkter nya funktioner uppstått och hur funktioner har förändrats under evolutionen. Vi undersöker i första hand genfamiljer som består av neuropeptider, G-proteinkopplade receptorer, jonkanaler och gener i synsystemet.
3) Att undersöka funktionellt de subtila egenskaperna hos G-proteinkopplade receptorer som avgör hur de kan binda och reagera på vissa ligander men ignorera andra. Vi fokuserar på adrenerga och dopaminerga GPCR eftersom deras naturliga ligander är mycket lika varandra, men ändå har de 11 adrenerga och de 9 dopaminerga receptorerna i ryggradsdjur (9 respektive 5 i däggdjur) vanligtvis tydliga ligand-preferenser. Vi utforskar receptorernas skillnader, som har stor farmakoterapeutisk betydelse, genom att använda evolutionära jämförelser, strukturmodellering, mutagenes, funktionell expression och farmakologisk karakterisering (bindning och signaltransduktion). Detta kommer att öka förståelsen av receptor-preferenser för många syntetiska agonister och antagonister som är kliniskt betydelsefulla. På sikt är målsättning att skräddarsy nya ligander och modifierade receptorer för att förbättra farmakologisk behandling av sjukdomar.
Många hundra, kanske tusentals, genfamiljer hos ryggradsdjur expanderade i två dramatiska händelser för ca 500 miljoner år sedan, nämligen två fördubblingar av arvsmassan (genomet), även kallade tetraploidiseringar. Dessutom inträffade en tredje tetraploidisering i föregångaren till de egentliga benfiskarna (teleoster). Dessa händelser förklarar en stor del av den genetiska och funktionella komplexiteten hos nutida ryggradsdjur och även varför olika medlemmar av samma genfamilj ofta har mer eller mindre överlappande funktioner. Vi använder en kombination av fylogenetiska sekvensanalyser och kromosomjämförelser mellan arter för att klarlägga genduplikationer i genfamiljer som är av särskilt intresse. Denna strategi är mycket användbar för att identifiera motsvarande gen (dvs ortolog gen) hos olika arter för jämförelse av funktioner. Resultaten har stor betydelse för våra möjligheter att förstå hur funktioner uppkommer och förändras och ibland även förloras under evolutionen. Bland de gener som vi undersökt, eller som vi för närvarande analyserar, finns peptiderna och receptorerna i neuropeptid Y-familjen, de opioida peptiderna (enkefaliner etc.) och deras receptorer, tillväxthormon och prolaktin och deras receptorer, oxytocin- och vasopressinreceptorer, somatostatinreceptorer, spänningsaktiverade natrium- kalcium- och kaliumkanaler, och receptorer för de viktiga neurotransmittorerna glutamat, acetylkolin, GABA, noradrenalin, dopamin och serotonin.
