Destruktionen och proliferationen av beta-celler.
Typ 1-diabetes är den vanligaste kroniska sjukdomen bland barn och ungdomar och på grund av de långsiktiga komplikationerna i samband med sjukdomen är den förväntade livslängden reducerad med mer än 10 år. Typ 1-diabetes utvecklas till följd av en progressiv förlust av insulinproducerande beta-celler orsakad av ett immunangrepp. Den underliggande utlösaren för immunsystemet är dock fortfarande okänd. Vid diagnos har 60-70% av betacellsmassan redan gått förlorad, baserat på obduktionsstudier. För närvarande finns det inga kliniskt validerade tekniker för att utvärdera beta-cells massa in vivo och därför är vår förståelse av hur beta-cells massan förändras begränsad till vad vi har lärt oss av obduktionsstudier. Genom att använda radioaktivt märkta positronemissionstomografi undersökningar (PET) som är specifika för beta-celler skulle det vara möjligt att bestämma beta-cells massan in vivo. PET är en mycket känslig, kvantitativ och icke-observatörsberoende medicinsk bildteknik där radiomärkta molekyler används för att spåra biologiska processer och receptorer. Min grupp fokuserar på utveckling av nya kliniskt tillämpliga tekniker för att bedöma beta-call massa och metoder som icke-invasivt kan avbilda och kvantifiera immunsvaret inom den mänskliga bukspottkörteln under utvecklingen av diabetes.
Vi arbetar också med att identifiera mekanismer involverade i destruktionen av beta-celler och nya läkemedel/ämnen med potential att öka proliferationen av beta-celler för att etablera regenerativa terapier för typ 1-diabetes. För detta ändamål tillämpar vi både experimentell in vitro- och in vivo-tekniker för att studera beta-cellers fysiologi.
